چکیده

We developed and validated a magnetizable solid-phase fluoroimmunoassayfor directlydeterminingquininein serum or urine. We prepared the Immunogen by coupling quinine hemisuccinate to keyhole-limpet hemocyanine, usingthis to immunizethree sheep, and couplingthe antiserato magnetizable solid-phaseparticlesto facilitateseparationof bound antigen from interferingcomponentsof serum or urine. We tested the stabilityof two fluorescein-labeledderivativesof quinine-one prepared by direct conjugationof quinine to dichlorotriazenylaminofluorescein,the other by conjugating quininehemisuccinateto fluoresceinthiocarbamylethylenediamine.The latterwas unstable.Usingthe formerlabeland an 30-mm incubation,we saw no interferenceby qumnidine (an enantiomerof quinine)or otherantimalarialdrugsat their therapeuticconcentrations.
